Venetola is an oral biologic that is a selective small molecule inhibitor of the anti apoptotic protein Bcl-2 and has potential anti-tumor activity. Venetola simulates the BH3 only protein, which is the native ligand and apoptosis activator of Bcl-2. By binding to the hydrophobic groove of Bcl-2 protein, it inhibits the activity of Bcl-2 and restores the apoptosis process of tumor cells. Bcl-2 protein is overexpressed in some cancers and plays an important role in regulating cell apoptosis; Its expression is related to the increase of drug resistance and tumor cell survival rate. Compared with the Bcl-2 inhibitor navitoclax, this drug does not inhibit bcl-XL and does not cause bcl-XL mediated thrombocytopenia.
